Abstract
Rekombinantni humani inzulin prvi je humani protein proizveden rekombinantnom DNA tehnologijom. Proces proizvodnje inzulina podrazumijeva sintezu gena, ugradnju gena u vektorsku molekulu, transformaciju stanice koja služi kao ekspresijski sustav i pročišćavanje dobivenog produkta. Standardni ekspresijski sustavi korišteni u industrijskoj proizvodnji su bakterija Escherichia coli i kvasac Saccharomyces cerevisiae. Rast broja dijabetičara i razvoj alternativnih načina terapije povećavaju potrebu za inzulinom koju trenutni mehanizmi proizvodnje ne mogu zadovoljiti. Intenzivno se istražuju složeniji ekspresijski sustavi (biljke i životinje) te mogućnost primjene tih sustava na proizvodnju velikih razmjera.
Abstract (english)
Recombinant human insulin was the first human protein made by recombinant DNA technology. The process of manufacturing insulin consists of gene synthesis, incorporation of the gene into a vector molecule, transformation of cells used as expression systems and finally protein purification. Standard expression systems in industrial protein manufacturing include the bacteria Escherichia coli and the yeast Saccharomyces cerevisiae. The incidence of diabetes is increasing rapidly along with alternative delivery systems development. The incerasing demand for insulin is barely met by current manufacturing capacities. More complex expression systems (plants, animals) and their large-scale manufacturing possibilities are being explored.
